This is the date of the microinjection session whose data you are describing in this row. Please, indicate which date format are you using (DD-MM-YYYY or MM-DD-YYYY) in the description of your facility. If you are using a Mac please read this help file regarding how Excel program encodes dates in different platforms before submitting your microinjection set of data. |
| CONSTRUCT |
Identification of the DNA construct used. Use numbers (i.e. 1, 2, 3, 4,... ) to differentiate data from different constructs. When injecting two or more constructs mixed together, please treat these as a new construct, even if one of them was injected by itself on a different date. If you provide real names of constructs, please be careful to always use the same name for the same construct. We will strip the real names and encode them to maintain confidentiality. |
| DNATYPE | The type of DNA construct that has been injected. Enter “1” for plasmid-based constructs and “2” for genomic-based (YAC, BAC, PAC) constructs. If you are co-microinjecting two plasmids please use “1”. If you are co-microinjecting two BACs please use “2”. |
| EMBRYO STRAIN | This is the strain of the embryos used for microinjection purposes. Please note that this may be different from the strain of females superovulated, if they are mated to a different strain of males, or if F1 hybrid males and females are mated (in the latter case, the embryos would be F2). Please use standard names to indicate the precise origin of the mice and, in particular, and whenever possible, please include your supplier/vendor's information. For example: for C57BL/6 mice, use C57BL/6J for the Jax substrain, C57BL/6NHsd for Harlan’s, C57BL/6NCrl for Charles River Laboratories’, and C57BL/6NTac for Taconic’s. |
| DONOR FEMALES SUPEROVULATED | This is the total number of oocyte donor females that were superovulated and from whom oviducts were harvested, irrespective of whether they were plugged or not by stud males. |
| EMBRYOS HARVESTED | This is the total number of oocytes that were collected from all donor females superovulated. This should include all oocytes having a normal appearance, irrespective of whether they were fertilised or not, or whether they could or couldn’t be microinjected. |
| EMBRYOS INJECTED | This is the total number of fertilised oocytes that were microinjected. |
| EMBRYOS TRANSFERRED | This is the total number of microinjected embryos that were eventually transferred to pseudopregnant females, either right after microinjection or after a variable incubation time. Normally, this number will reflect the loss of some eggs due to the mechanical trauma of injection. If some eggs were not implanted for other reasons known to you, please state these reasons in the "Incidences" column. |
| PSEUDOPREGNANT FEMALES USED | This is the total number of pseudopregnant females to which the embryos were transferred, irrespective of whether they became pregnant or not. |
| MOUSE STRAIN FOR PSEUDOS | This is the mouse strain being used for pseudopregnant females. Please use standard names to indicate the precise origin of the mice. For example: for CD-1 outbred mice, use Hsd:ICR(CD-1) for Harlan’s, Crl:(CD-1)ICR for Charles River Laboratories’, and IcrTac:ICR for Taconic’s. |
| TOTAL PUPS | This is the total number of pups delivered, reaching to term, irrespective of whether they were alive or dead. This number also includes the number of fetuses to term that might have been delivered by caesarean section. No reabsorbed embryos should be included here. |
| TRANSGENIC PUPS | This is the total number of transgenic mice (DNA positive by PCR and/or Southern blot DNA analysis) detected among the obtained total pups. |
| INCIDENCES | This is a free-text field. Please use it to report any incidence that might have happened during this session that you think could have influenced the outcome of the microinjection session. For example “first days of microinjection by this person”, “new microscope or injector bought”, “bad batch of hormones”, “construction in the animal house”, “embryos lost in the incubator”, “different persons carrying out the work”, or, if you wish to take into account different people working at your facility you can include "microinjection done by person 1", "microinjection done by person 2" , ...etc. |
